摘要:
摘 要:目的 将炙甘草的化学成分与网络药理学相结合,探讨炙甘草抗心律失常的作用机理。方法 运用UPLC-TOF-MS、GC-MS技术分析炙甘草的化学成分。利用TCMSP数据库筛选活性成分,通过Swiss Target Prediction数据库和Gene Cards数据库搜集药物作用靶点和疾病靶点,借助Cytoscape 3.8.2软件筛选核心靶点,对核心靶点进行蛋白互作网络分析,GO和KEGG富集分析,使用Cytoscape 3.8.2构建药物关键成分核心靶点通路图,分子对接验证由Autodocking vina 1.2.0软件完成。结果 共分析出289种化学成分,其中异甘草素、酪氨酸等16个成分为抗心律失常的关键成分;蛋白互作网络共筛选出ALB、AKT1、VEGFA、EGFR等55个核心靶点,GO富集共获得476个条目,KEGG共得到130条通路。分子对接结果表明,异甘草素、黄豆黄素、蜜桔黄素与核心靶点对接活性较好。结论 炙甘草可能通过调节ALB、AKT1、VEGFA、EGFR等多个靶点,以及参与癌症通路、癌症中的蛋白聚糖、内分泌耐药、EGFR酪氨酸激酶抑制剂耐药等信号通路发挥抗心律失常的作用。
Abstract:
Abstract: Objective To combine the chemical components of roasted licorice with network pharmacology and explore the mechanism of its anti arrhythmia effect.Methods UPLC-TOF-MS and GC-MS techniques were used to analyze the chemical components of roasted licorice.TCMSP database was used to screen active ingredients,Swiss Target Prediction database and Gene Cards database were used to collect drug action targets and disease targets,Cytoscape 3.8.2 software was used to screen core targets,and protein interaction network analysis,GO and KEGG enrichment analysis were carried out on core targets.Cytoscape 3.8.2 was used to construct drug key component core target pathway diagram,and Macromolecular docking verification was completed by Autodocking vina 1.2.0 software.Results A total of 289 chemical components were analyzed,among which 16 components such as isoliquiritigenin and tyrosine were key components for anti arrhythmias;The protein interaction network screened 55 core targets,including ALB,AKT1,VEGFA,EGFR,etc.GO enrichment resulted in a total of 476 entries,and KEGG resulted in a total of 130 pathways.The results of Macromolecular docking showed that isoliquiritigenin,daidzein and citrinin had good docking activity with the core target.Conclusion Roasted licorice may play an antiarrhythmic role by regulating multiple targets such as ALB,AKT1,VEGFA,and EGFR,as well as participating in cancer pathways,Proteoglycan in cancer,endocrine resistance,and resistance to EGFR Tyrosine kinase inhibitor.
