摘要:
摘?要：目的 观察金果胃康胶囊对胃癌前病变大鼠胃黏膜HSP27、MAD2蛋白表达的影响。方法 将85只大鼠随机分为空白组10只和造模组75只。空白组给予正常饮食，造模组采用N-甲基-N-硝基-N-亚硝基胍(MNNG)联合乙醇、氨水及饥饱失常组成复合造模法。将造模后大鼠随机分成模型组、金果胃康胶囊高、中、低剂量组和维酶素组。各给药组分别给予相应药物灌胃，空白组和模型组给予等量生理盐水灌胃，1次·d-1，连续12 w。12 w后处死所有大鼠，取胃体组织，光镜下观察各组大鼠胃黏膜组织病理变化；免疫组化法检测胃黏膜组织中HSP27、MAD2蛋白表达情况。结果 模型组大鼠HSP27、MAD2蛋白表达水平明显高于空白组(均P＜005)。各给药组HSP27、MAD2蛋白表达水平明显低于模型组(P均＜0.05)，且金果胃康胶囊中高剂量组明显低于维霉素组(均P＜0.05)。结论 金果胃康胶囊可逆转胃癌前病变的发生，作用机制可能与其能抑制HSP27蛋白的表达及控制MAD2蛋白的表达有关。
Abstract:
Abstract： Objective To observe the effect of Jinguo Weikang Capsule on the expression of HSP27 and MAD2 proteins in the gastric mucosa of rats with gastric precancerous lesions.Methods 85 rats were randomly divided into a blank group of 10 and a model group of 75.The blank group was given a normal diet,while the modeling group used a composite modeling method consisting of Nmethyl-N-nitro-Nnitrosoguanidine (MNNG),ethanol,ammonia,and hunger and satiety disorders.Randomly divide the rats after modeling into model group,high,medium,and low dose groups of Jinguo Weikang capsules,and vitamin enzyme group.Each medication group was given corresponding drugs by gavage,while the blank group and model group were given equal amounts of physiological saline by gavage once per day for 12 consecutive weeks.After 12 weeks,all rats were euthanized,and gastric tissue was taken.Pathological changes in the gastric mucosa tissue of each group of rats were observed under light microscopy;Immunohistochemical method was used to detect the expression of HSP27 and MAD2 proteins in gastric mucosal tissue.Results The expression levels of HSP27 and MAD2 proteins in the model group were significantly higher than those in the blank group (all P<0.05).The expression levels of HSP27 and MAD2 proteins in each treatment group were significantly lower than those in the model group (all P<0.05),and the highdose group of Jinguo Weikang Capsule was significantly lower than the Weimycin group (all P<0.05).Conclusion Jinguo Weikang Capsule can reverse the occurrence of gastric precancerous lesions,and the mechanism of action may be related to its ability to inhibit the expression of HSP27 protein and control the expression of MAD2 protein.
