摘?要:目的 研究荔枝核总黄酮(Total flavonoids from Litchi seed,TFL)对猪血清诱导的大鼠免疫性肝纤维化的影响及其可能的分子学机制。方法 将60只SD大鼠,分为正常对照组、肝纤维化模型组、TFL低、中、高剂量组及秋水仙碱阳性对照组,每组10只。实验通过腹腔注射猪血清(每次注射0.5 mL,2次·w-1,共12 w)构建大鼠免疫性肝纤维化模型,正常对照组不造模,造模同时,TFL低剂量组(100 mg·kg-1 TFL混悬)、中剂量组(200 mg·kg-1 TFL混悬)、高剂量组(300 mg·kg-1 TFL混悬)及秋水仙碱阳性对照组(0.5 mg·kg-1)分别给予相应剂量的受试药物灌胃,第12 w后处死大鼠,取肝组织HE染色,ELISA法检测大鼠血清中透明质酸(hyaluronic acid,HA)、Ⅲ型前胶原(type Ⅲ procollagen,PCⅢ)、层粘连蛋白(laminin,LN)和Ⅳ型胶原(type Ⅳ collagen,CⅣ)的水平;Western blot检测大鼠肝脏织早期生长反应因子(early growth response factor,EGR1)、转化生长因子β1(transforming growth factor β1,TGF-β1)蛋白表达情况。结果 与正常对照组比较,模型组大鼠血清中HA、PCⅢ、LN及CⅣ的含量明显升高(P<0.05),肝组织匀浆中EGR1、TGFβ1蛋白表达显著升高(P<0.05),肝脏病理学提示肝纤维评分(4.90±0.57)明显加重(P<0.05);与模型组比较,TFL高、中、低剂量组大鼠血清中HA、PCⅢ、LN及CⅣ的含量明显降低(P<0.05),肝组织匀浆中EGR1、TGF-β1蛋白表达显著降低(P<0.05),并且肝纤维化程度减轻(P<0.05)。结论 TFL可能通过降低肝组织EGR1、TGF-β1蛋白的表达减轻猪血清诱导的免疫性肝纤维化。
Abstract:
Abstract: Objective To investigate the effect of Total flavonoids from Litchi seed (TFL) on immune liver fibrosis induced by pig serum in rats and its possible molecular mechanisms.Methods Sixty SD rats were divided into normal control group,liver fibrosis model group,low,medium,and high dose TFL groups,and colchicine positive control group,with 10 rats in each group.The experimental model of immune liver fibrosis in rats was established by intraperitoneal injection of pig serum (0.5 mL,2 times·w-1,for a total of 12 weeks).The normal control group did not make the model,and at the same time,the low dose group (100 mg·kg-1 TFL suspension),medium dose group (200 mg·kg-1 TFL suspension),high dose group (300 mg·kg-1 TFL suspension),and colchicine positive control group (0.5 mg·kg-1) were given corresponding doses of the test drug by gavage,After the 12th week,the rats were euthanized and their liver tissue was stained with HE.The levels of hyaluronic acid (HA),type Ⅲ procollagen (PC Ⅲ),laminin (LN),and type Ⅳ collagen (cⅣ) in the rat serum were measured using ELISA;Western blot detection was applied to detect the protein expression of early growth response factor (EGR1) and transforming growth factor(TGF-β1) in rat liver tissue.Results Compared with the normal control group,the content of HA,PC Ⅲ,LN,and cⅣ in the serum of the model group rats was significantly increased (P<0.05),and the expression of EGR1 and TGF-β1 proteins in the liver tissue homogenate was significantly increased (P<0.05).Liver pathology showed a significant increase in liver fiber score (P<0.05);Compared with the model group,the levels of HA,PC Ⅲ,LN,and cIV in serum of rats in the high,medium,and low dose TFL groups were significantly reduced (P<0.05),and the expression of EGR1 and TGF-β1 proteins in liver tissue homogenate was significantly reduced (P<0.05),and the degree of liver fibrosis was reduced (P<0.05).Conclusion TFL may alleviate immune liver fibrosis induced by pig serum by reducing the expression of EGR1 and TGF-β1 proteins in liver tissue.
