摘要:
摘 要:目的 探讨甘草泻心汤联合血竭对硫酸葡聚糖钠(DSS)诱导的溃疡性结肠炎(UC)的抗炎作用。方法 SPF级SD大鼠100只按随机数字表法分为25只空白组和75只造模组,造模组大鼠采用3% DSS自由饮用法造模,造模成功后大鼠分为模型组、柳氮磺胺吡啶(SASP)组、甘草泻心汤+血竭组各25只,甘草泻心汤+血竭组每天给予甘草泻心汤6 g?(kg?d)-1、血竭375 mg?(kg?d)-1混悬液灌胃,柳氮磺吡啶组每天给予柳氮磺吡啶450 mg?(kg?d)-1混悬液灌胃,空白组和模型组每天给予等体积生理盐水2.5 mL /只灌胃,连续灌胃7 d。观察大鼠一般情况及结肠HE染色后炎症程度,对大鼠进行疾病活动指数(DAI)、结肠黏膜损伤(CMDI)及病理组织学(HS)评分,ELISA法检测大鼠血清白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)的含量。结果 模型组大鼠体质量低于空白组,DAI、CMDI、HS评分及血清IL6、TNF-α含量均高于空白组,差异有统计学意义(P<0.05);甘草泻心汤+血竭组和柳氮磺吡啶组大鼠体质量高于模型组,DAI、CMDI、HS评分及血清IL-6、TNF-α含量均低于模型组,差异有统计学意义(P<0.05);甘草泻心汤+血竭组HS评分及血清IL-6、TNF-α含量低于柳氮磺吡啶组,差异有统计学意义(P<0.05);甘草泻心汤+血竭组与柳氮磺吡啶组间大鼠体质量、DAI、CMDI评分差异无统计学意义(P>0.05)。结论 甘草泻心汤联合血竭灌胃能改善核酸葡聚糖钠(dextran sulfate sodium,DSS)诱导的UC模型大鼠症状,降低UC大鼠结肠DAI、CMDI及HS评分,抑制UC大鼠血清IL-6、TNF-α的表达,具有明确的抗溃疡性结肠炎作用。
Abstract:
Abstract: Objective To investigate the antiinflammatory effect of Gancao Xiexin decoction Combined with Dragon's blood on ulcerative colitis (UC) induced by dextran sulfate sodium (DSS).Methods 100 SPF SD rats were randomly divided into 25 blank groups and 75 model groups.The model rats were made by 3% DSS free drinking method.After successful modeling,the rats were divided into model group,sulfasalazine (SASP) group and Gancao Xiexin decoction + Dragon's blood group,with 25 rats in each group.The Gancao Xiexin decoction + Dragon's blood group was given Gancao Xiexin decoction 6 g?(kg?d)-1 Dragon's blood 375 mg ? (kg ? d)-1 suspension was administered by gavage,sulfasalazine group 450 mg ? (kg ? d)-1 suspension was administered by gavage every day,blank group and model group were administered by gavage of equal volume normal saline 25 mL?case-1 every day for 7 days.The general condition of rats and the degree of inflammation after colonic HE staining were observed.The rats were scored by disease activity index (DAI),colonic mucosal injury (CMDI) and histopathology (HS).The serum content ofinterleukin6 (IL-6) and tumor necrosis factor α( TNF-α) were detected by ELISA .Results The body mass of the model group was lower than that of the blank group,DAI,CMDI,HS scores and serum IL6 and TNFα.The body mass of rats in Gancao Xiexin decoction + Dragon's blood group and sulfasalazine group was higher than that in model group,DAI,CMDI,HS scores,serum IL6 and TNFα The content of HS score and serum IL6 and TNFα in Gancao Xiexin decoction + Dragon's blood group were significantly lower than those in model group (P < 0.05).The content was lower than that of sulfasalazine group.Conclusion Gavage of Gancao Xiexin decoction Combined with Dragon's blood can improve the symptoms of DSS induced UC model rats,reduce the colon DAI,CMDI and HS scores of UC rats,and inhibit the serum IL6 and TNFα of UC rats.It has a clear anti ulcerative colitis effect.
