摘要:
目的 研究灯心草提取物(JE)对异丙肾上腺素(ISO)诱导的大鼠心肌纤维化的影响并探讨相关机制。 方法 将40只SD大鼠随机分成5组,每组8只,分为对照组、模型组、低(90mg.kg-1.d-1)、中(180mg.kg-1.d-1)、高(360mg.kg-1.d-1)剂量JE组。模型组和JE组每天背部皮下注射ISO(5mg.kg-1.d-1),持续7天建立大鼠MF模型,对照组则予等量0.9% 生理盐水。 自造模成功第二天起,低、中、高剂量JE组分别给予 不同剂量JE腹腔注射21天,对照组和模型组注射等量生理盐水。 通过HE和Masson 染色观察心肌纤维化程度 并计算胶原容积分数(CVF);ELISA 检测大鼠心肌组织中Ⅰ型胶原(ColI)、Ⅲ型胶原(ColⅢ)含量;Western blot 检测大鼠心肌组织中 TGF-β1及SMAD2/3 蛋白表达。 结果 模型组与对照组相比,心肌细胞排列较乱并纤维 化,有大量胶原纤维沉积,CVF 显著升高(P<0.01);与模型组相比,低剂量JE组MF有不同程度改善,随着剂量 逐渐增加,纤维排列规律,胶原沉积显著减少,中、高剂量 JE 组的表现更加明显,CVF水平显著降低(P<0.05), 这与JE剂量的增加明显相关。 与对照组比较,模型组中 ColI 和 ColⅢ含量明显增加(P<0.01);与模型组对比, 低、中、高JE组的大鼠心肌组织中 ColI 和 ColⅢ含量均降低(P<0.01);随着JE剂量增加,其下降幅度越大,高 剂量JE组的效果最显著(P<0.05)。 与对照组相比,模型组 TGF-β1及SMAD2/3 蛋白表达明显升高(P<0.01);与模型组比较,低、中、高剂量JE组 TGF-β1及SMAD2/3 蛋白表达呈显著下降趋势(P<0.05);随着JE剂量增加,下降趋势越明显,高剂量JE组的效果更显著(P<0.05)。 结论 灯心草可以改善ISO诱导的大鼠心肌 纤维化,随着JE剂量增加其效果更加明显,其机制可能与JE剂量下 TGF-β1及SMAD2/3蛋白表达的变化有关。
Abstract:
Objective: To study the effects of Herba rushanensis extract ( JE) on isoproterenol ( ISO) - induced myocardial fibrosis in rats and to explore the related mechanism. Methods: Forty SD rats were randomly divided into 5 groups, 8 in each group. They were divided into control group, model group, low (90 mg.kg - 1.D - 1 ), medium (180 mg.kg - 1.D - 1 ) and high (360 mg.kg - 1.D - 1 ) JE groups. Rats in model group and JE group were subcutaneously injected with ISO (5 mg.kg - 1.D - 1 ) every day for 7 days to establish MF model, while the control group was given equal amount of 0. 9% normal saline. From the second day after successful modeling, low, medium and high dose JE groups were given different doses of JE intraperitoneal injection for 21 days, and the control group and model group were injected with the same amount of normal saline. The degree of myocardial fibrosis was observed by HE and Masson staining, and the collagen volume fraction (CVF) was calculated. The contents of collagen type Ⅰ (ColI) and collagen type Ⅲ (Col Ⅲ) were detected by ELISA. The expression of TGF - β 1 and Smad2 / 3 protein in myocardial tissue was detected by Western blot. Results: Compared with the control group, the myocardial cells in the model group were disorderly arranged and fibrosis, with a large amount of collagen fiber deposition, and CVF was significantly increased (P < 0. 01); compared with the model group, MF in the low - dose JE group was improved in varying degrees, with the gradual increase of the dose, the fiber arrangement and collagen deposition were significantly reduced, and the performance of the medium and high dose JE groups was more obvious, and the CVF level was significantly decreased (P < 0. 05), This was significantly related to the increase of JE dose. Compared with the control group, the contents of ColI and Col Ⅲ in the model group were significantly increased (P < 0. 01); compared with the model group, the contents of ColI and Col Ⅲ in the myocardial tissue of rats in the low, medium and high JE groups were decreased (P < 0. 01); with the increase of JE dosage, the greater the decrease range was, and the effects of high dose JE group was the most significant (P < 0. 05). Compared with the control group, the expression of TGF - β 1 and Smad2 / 3 protein in the model group was significantly increased (P < 0. 01); compared with the model group, the expression of TGF - β 1 and Smad2 / 3 protein in the low, medium and high dose JE groups showed a significant downward trend (P < 0. 05); with the increase of JE dose, the downward trend became more obvious, and the effects of high dose JE group was more significant (P < 0. 05). Conclusion: Herba Leonuri can improve myocardial fibrosis induced by ISO in rats, and the effect is more obvious with the increase of JE dose. The mechanism may be related to the changes of TGF - β 1 and Smad2 / 3 protein expression.
