摘要:
观察桂枝茯苓胶囊对子宫内膜异位症(EMs)大鼠异位内膜组织中水通道蛋白 1、2、5(AQP1、 2、5)基因表达的影响。方法 随机将 EMs模型大鼠分为模型组、桂枝茯苓胶囊高、低剂量组、散结镇痛胶囊组、丹 莪妇康煎膏组,每组15只。另设正常对照组15只。用桂枝茯苓胶囊、散结镇痛胶囊、丹莪妇康煎膏对 EMs大鼠 进行干预,给药 4周后,实时荧光定量 PCR(RT-qPCR)法检测 EMs大鼠异位内膜 AQP1、2、5mRNA的表达量。 结果 与正常对照组相比,模型组异位内膜 AQP1mRNA表达显著升高,模型组异位内膜 AQP2、AQP5mRNA表达 显著下降;与模型组相比,桂高组异位内膜 AQP1mRNA表达显著下降,桂高组异位内膜 AQP2、AQP5mRNA表达 显著升高。结论 桂枝茯苓胶囊治疗 EMs的机制与下调大鼠异位内膜组织中 AQP1的基因表达,上调 AQP2、 AQP5的基因表达有关,进而抑制异位内膜血管生长,使异位内膜萎缩、坏
Abstract:
Objective: To observe the effect of Guizhi Fuling Capsule on the expression of aquaporin 1, 2, 5 ( AQP1,2, 5) in ectopic endometrium of endometriosis ( EMs) rats. Methods: The EMs model rats were randomly divided into model group, Guizhi Fuling capsule high and low dose group, Sanjie Zhentong capsule group and Danqi Fukang ointment group, with 15 rats in each group. Another 15 rats in the normal control group were set up. EMs rats were treated with Guizhi Fuling Capsule, Sanjie Zhentong Capsule and Danqi Fukang Decoction. After 4 weeks of administration, the expression of AQP1, 2 and 5 in ectopic endometrium of EMs rats was detected by real - time fluorescence quantitative PCR ( RT - qPCR) . Results: Compared with the normal control group, the expression of AQP1 mRNA in the ectopic endometrium of the model group was significantly increased, and the expression of AQP2 mRNA and AQP5 mRNA in the ectopic endometrium of the model group was significantly decreased. Compared with the model group, the expression of AQP1 mRNA in the ectopic endometrium of the Guigao group was significantly decreased. The expression of AQP2 and AQP5 mRNA in ectopic endometrium of Guigao group was significantly increased. Conclusion: The mechanism of Guizhi Fuling Capsule in treating EMs is related to down - regulation of AQP1 gene expression in ectopic endometrium and up - regulation of AQP2 and AQP5 gene expression in ectopic endometrium of rats, which inhibits the growth of ectopic endometrium vessels and causes atrophy and necrosis of ectopic endometrium.
