摘要:
目的 比较滋补肾阴法、温补肾阳法对 ＨＢＶ 免疫耐受患者 ＤＣｓ 功能影响。 方法 采集 ２０ 例 ＨＢＶ 免疫耐受患者和 １０ 例健康者外周血，免疫磁珠法分离单个核细胞（ＰＢＭＣｓ），分离提取、培养 ＤＣ 细胞，采用滋补肾阴、温补肾阳药物制备的大鼠含药血浆进行培养干预，检测 ＤＣｓ 表面 ＣＤ８０、ＣＤ８６、ＨＬＡ － ＤＲ、ＩＬ － １２ 水平。 结果药物血浆处理免疫耐受组 ＤＣｓ 后 ＣＤ８０、ＣＤ８６、ＨＬＡ － ＤＲ、ＩＬ － １２ 表达率升高，以 ＣＤ８０、ＩＬ － １２ 升高最为明显（ｐ ＜ ０． ０５），温补肾阳药物血浆优于滋补肾阴组药物血浆和桂附汤血浆，六味地黄组优于桂附组。 结论 温补肾 阳法和滋补肾阴法含药血浆均能提高 ＨＢＶ 免疫耐受患者来源的 ＤＣｓ 细胞功能，温补肾阳法对 ＨＢＶ 免疫耐受患者 ＤＣｓ 功能有着更显著的影响。
Abstract:
To compare the effects of nourishing kidney - yin method and warming kidney - yang method on DCs function in patients with HBV immune tolerance. Method Peripheral blood was collected from 20 patients with HBV immune tolerance and 10 healthy persons. Mononuclear cells ( PBMCs) were isolated by immunomagnetic bead method. DC cells were isolated, extracted and cultured. Rat plasma containing drugs for nourishing kidney yin and warming kidney Yang was used for culture intervention. CD80, CD86, HLA - DR and IL -12 levels on the surface of DCs were detected. Result The expression rates of CD80, CD86, HLA - DR and IL - 12 in DCs treated with drug plasma were increased, especially in CD80 and IL -12 ( p <0. 05) . The plasma of warming and tonifying kidney - yang drug was better than that of nourishing kidney - yin drug and Guifu decoction, and that of Liuwei Dihuang group was better than that of Guifu group. Conclusion Both the methods of warming and tonifying kidney - yang and nourishing kidney - yin canimprove the function of DC cells from patients with HBV immune tolerance, and the method of warming and tonifying kidney - yang has more significant effect on DCs function of patients with HBV immune tolerance.
