摘?要:目的 利用网络药理学、分子对接技术探究金银花抗流感的有效成分和作用机制。方法 采用数据库与文献检索收集金银花有效成分与其作用靶点,通过分子对接、靶点预测构建药物-靶点(D-T)网络、药物-靶点-疾病(D-T-D)网络、GO和KEGG富集性分析,阐明金银花分子与靶点和疾病的关系。结果 共获取金银花活性成分32个,对应靶点56个;分子对接分析金银花可直接作用于流感病毒表面蛋白血凝素(Hemagglutinin,HA)与神经氨酸酶(neuramidinase,NA)及其突变型,还可作用受体MMP12、MMP9、MMP2、ALDH2、ALOX5、OPRM1、OPRD1、OPRK1、ADRB2而对呼吸系统疾病、发热及炎症进行干预,GO富集分析得到生物学过程148条、分子功能37条以及细胞组分45条,主要与炎症反应、G蛋白偶联受体以及MAPK激活过程相关;KEGG富集分析得出23条显著富集的通路,主要有TNF信号通路、钙信号通路、花生四烯酸代谢通路等。结论 金银花能够通过多化合物作用于多个靶点发挥抗流感作用,这其中包括对流感病毒及其突变型的直接作用和通过干预流感病毒导致的呼吸系统疾病、发热和炎症症状两个方面,其分子机制主要从系统水平上调节多种生物过程参与炎症和呼吸系统疾病的治疗。
