摘 要:目的 探讨足三里防治非甾体抗炎药胃溃疡的作用机理。方法 SD大鼠按随机数字法分为正常组、模型组、足三里组,每组8只。模型组、足三里组采用腹腔注射吲哚美辛制备胃溃疡模型:足三里组采用电针预处理结合穴位注射NS(生理盐水)干预,干预方式:造模前6 d,连续电针大鼠双侧足三里20 min,1次/d,并于第7 d造模后60 min,于双侧足三里注射NS 0.5 mL·kg-1;各组大鼠于造模后7 h麻醉,取血清、胃,计算胃黏膜损伤分数、HE染色观察胃黏膜病理形态。检测谷胱甘肽过氧化物酶(GSHPx)活力,表皮生长因子(EGF)、胃泌素(Gas)含量,以及胃Nrf2、GSTm1基因表达。结果 与模型组比,足三里组胃黏膜大体外观及病理形态损伤明显改善,胃黏膜损伤分数显著降低(P<0.01);与正常组比,模型组大鼠胃组织GSHPx活力,胃组织和血清中EGF含量下降(P均<0.01)、Gas升高(P<0.05、P<0.01);与模型组比,足三里组大鼠胃组织GSHPx活力、胃组织和血清EGF含量均升高(P<0.05、P<0.01)、Gas均明显下降(P<0.01);与正常组比,模型组大鼠胃组织GSTm1 mRNA表达下降(P<0.05);与模型组比,足三里组大鼠胃组织GSTm1 mRNA表达上升(P<0.05)。结论 足三里保护吲哚美辛实验性胃溃疡大鼠胃黏膜的作用,与提高胃组织GSHPx活力、促进EGF分泌,降低胃泌素分泌,促进胃组织GSTm1 mRNA表达有关,针灸可能在抗氧化、促进胃黏膜修复、降低胃酸分泌方面发挥防治胃溃疡作用。
Abstract:
Abstract:Objective To investigate the mechanism of Zusanli in preventing and treating gastric ulcer with NSAID.Methods SD rats were randomly divided into normal group,model group and Zusanli group with 8 rats in each group.The model group and Zusanli group were treated with intraperitoneal injection of indomethacin to prepare gastric ulcer model.The Zusanli group was treated with electroacupuncture pretreatment combined with acupoint injection of NS (normal saline).The intervention mode was:6 days before modeling,the rats were continuously electroacupunctured at Zusanli on both sides for 20 minutes,once a day,and at 60 minutes after modeling on the 7th day,NS 0.5 mL·kg-1 was injected at Zusanli on both sides;The rats in each group were anesthetized 7 hours after modeling,the serum and stomach were taken,and the gastric mucosa injury fraction was calculated,and the pathological morphology of gastric mucosa was observed by HE staining.The activity of glutathione peroxidase (GSH Px),the content of epidermal growth factor (EGF) and gastrin (Gas),and the expression of Nrf2 and GSTm1 genes in stomach were detected.Results Compared with the model group,the gross appearance and pathological morphology of gastric mucosa in Zusanli group were significantly improved,and the score of gastric mucosa damage was significantly reduced (P<0.01);Compared with the normal group,the activity of GSH Px in gastric tissue,the content of EGF in gastric tissue and serum in the model group decreased (P<0.01),and the content of gas increased (P<0.05,P<0.01);Compared with the model group,the activity of GSH Px in stomach tissue,the content of EGF in stomach tissue and serum of rats in Zusanli group increased (P<0.05,P<0.01),and the content of gas decreased significantly (P<0.01);Compared with the normal group,the expression of GSTm1 mRNA in the stomach tissue of the model group decreased (P<0.05);Compared with the model group,the expression of GSTm1 mRNA in stomach tissue of rats in Zusanli group increased (P<0.05).Conclusion Zusanli protects the gastric mucosa of indomethacin induced experimental gastric ulcer rats,which is related to the increase of GSH Px activity in gastric tissue,the promotion of EGF secretion,the reduction of gastrin secretion,and the promotion of GSTm1 mRNA expression in gastric tissue.Acupuncture and moxibustion may play a role in preventing and treating gastric ulcer in terms of antioxidation,the promotion of gastric mucosal repair,and the reduction of gastric acid
