摘要:
目的探索脾虚痰湿型肥胖大鼠模型的制备方法?方法24只SD雄性大鼠按体重随机分为正常组(n=8)?模型A组(n=8)?模型B组(n=8)?模型A?B两组采用不同的干预顺序,观察各组大鼠一般行为学的变化情况及毛色?大便?体质量?精神状态?计算脾虚积分;测定大鼠总胆固醇(TC)?甘油三脂(TG)?高密度脂蛋白胆固醇(HDL-C)?低密度脂蛋白胆固醇(LDL-C)?谷丙转氨酶(ALT)并观察肝脏病理变化?结果经过高脂饲料喂养,模型组体重超过正常组(P<0.05);经过造模干预后模型组大鼠形体肥胖?被毛油腻?活动迟缓?倦怠蜷卧?大便稀软,符合脾虚痰湿的一般表现;模型A?B组均有不同程度的脂肪肝,且模型A组大鼠出现肝功能损伤较明显?结论通过高脂饲料喂养+饮食不节?劳倦过度?寒凉攻下的多因素造模法能够制备出脾虚痰湿型肥胖大鼠,但仍有些许地方值得改进;且模型A组较模型B组的方法更为满足模型要求,符合预期结果?
Abstract:
Objective To explore preparing method of obese rat model with spleen deficiency and phlegm dampness. Methods Twenty - four SD male rats were randomly divided into normal group ( n =8) , model A group ( n =8) and model B group ( n =8) . Models A and B were treated with different intervention sequences. The general behavioral changes and hair color, stool, body mass, mental state, and spleen deficiency scores were observed. Total cholesterol ( TC) , triglyceride ( TG) , high density lipoprotein cholesterol ( HDL - C) , low density lipoprotein cholesterol ( LDL - C) , glutathione transaminase ( ALT) were measured and liver pathology was observed. Results After feeding with high - fat diet,the weight of model group was higher than that of normal group ( P < 0. 05) ; after intervention, the model group rats were obese with greasy coat, sluggish activity and loose stool, easy to get tired and lying down, which accorded with the general manifestation of spleen deficiency and phlegm dampness; the model group A and B all had different degrees of fatty liver, and the liver function damage of model group A rats was more obvious. Conclusion The obese rats with spleen deficiency and phlegm dampness can be prepared by feeding with high fat diet, excessive fatigue and cold attack, but there are still some places to be improved; and the method of model group A is more satisfied with the model requirementsthan that of model group B, and meets the expected results.
