摘要:
目的 观察半夏泻心汤及其拆方对溃疡性结肠炎( UC) 模型大鼠结肠组织细胞因子含量变化的影 响,探讨半夏泻心汤及其拆方治疗 UC 的作用机制。方法 以 SD 大鼠为研究对象,采用 TNBS 乙醇灌肠法复制溃 疡性结肠炎动物模型,通过半夏泻心汤及其拆方给药,双抗体一步夹心酶联免疫法检测结肠组织 TNF - α、 IL - 1、 IL -10、 PGE2、 MPO 含量; HE 染色行病理组织学观察。结果 与模型组比较,半夏泻心汤组、寒热并用组( 半 夏、干姜、黄芩、黄连) 、辛温组( 半夏、干姜) 、甘调组( 人参、炙甘草、大枣) 和苦降组( 黄芩、黄连) TNF - α、 IL - 1 和 MPO 含量均降低( P <0. 05) ;与半夏泻心汤组比较,寒热并用组、辛温组、甘调组、苦降组 TNF - α、 IL -1 含量 较高( P 0. 05) ,辛温组、甘调组 MPO 含量较高( P < 0. 05) ; 与寒热并用组比较,辛温组和苦降组 TNF - α、 IL - 1 含量较高( P <0. 05) ,辛温组 MPO 含量较高( P <0. 05) 。结论 半夏泻心汤及其拆方对 UC 大鼠结肠组织 TNF - α、 IL -1、 MPO、 PGE2 影响最明显的是半夏泻心汤组,其次是寒热并用组、辛温组、苦降组、甘调组;半夏泻心汤及 其拆方对 UC 大鼠结肠组织病理学改变影响最明显的是半夏泻心汤组和寒热并用组,其次是苦降组和辛温组。
Abstract:
To observe the effect of Banxia Xiexin Decoction and its decomposed formula on the changes of cytokine content in the colon tissue of ulcerative colitis ( UC) rats,and to explore the mechanism of the effect of Banxia Xiexin Decoction and its disassembled prescription in the treatment of UC. Method: SD rats were used as the research object,and the animal model of ulcerative colitis was replicated by TNBS ethanol enema method. The contents of TNF -alpha,IL -1, IL -10,PGE2 and MPO in colonic tissue were detected by double antibody and sandwich enzyme - linked immunosorbent assay. Histopathological observation was carried out by HE staining. Result: Compared with the model group, the content of TNF - a, IL -1 and MPO in Banxia Xiexin Decoction group,cold and warm natured group ( pinellia ternata,dried ginger,scutellaria,rhizoma coptidis) ,pungent and warm natured group ( pinellia ternata,dried ginger) and sweet nursing group ( radix ginseng,radix glycyrrhizae,jujube) and bitter descending group ( scutellaria baicalensis,rhizoma coptidis) all decreased ( P < 0. 05) . Compared with Banxia Xiexin Decoction group,the content of TNF - a,IL -1 in cold and warm natured group,pungent and warm natured group,sweet nursing group and bitter descending group were relatively high ( P 0. 05) , the content of MPO in pungent and warm natured group and sweet nursing group were relatively high ( P <0. 05) . Compared with cold and warm natured group, the content of TNF - a,IL -1 in in pungent and warm natured group and bitter descending group were relatively high ( P <0. 05) , the content of MPO in pungent and warm natured group was relatively high ( P <0. 05) . Conclusion: The effect of Banxia Xiexin Decoction and its decomposed formula on TNF - α,IL -1,MPO and PGE2 in colon tissue of UC rats was the most obvious in Banxia Xiexin Decoction group, followed by cold and warm natured group,pungent and warm natured group,bitter descending group and sweet nursing group. Banxia Xiexin Decoction and its decomposed formula on the pathological changes of colon tissue in UC rats were most obvious in the Banxia Xiexin Decoction group and cold and warm natured group, followed by bitter descending group and pungent and warm natured group.
