摘要:
观察葛根通络颗粒的镇痛作用及其机制。方法 将 48 只筛选合格小鼠随机分为: 模型组、葛 根通络颗粒低、中、高剂量组( 3. 0、6. 0、12. 0g·kg- 1 ,以生药计,以下简称 GG 低组、GG 中组、GG 高组) ,模型组 仅给予 0. 5% CMC - Na 混悬液( 0. 2mL/10g) ,采用热板法观察各组小鼠痛阈值变化情况; 将 50 只筛选合格小鼠 随机分为模型组、GG 低组、GG 中组、GG 高组和阿司匹林组( 0. 2g/kg) ,模型组仅给予 0. 5% CMC - Na 混悬液 ( 0. 2mL/10g) ,各组小鼠均在给药后 1 小时按 0. 1mL/10g 腹腔注射 0. 6% 冰醋酸( HAC) ,观察 15min 后各组小鼠 扭体反应次数,记录次数并计算抑制率; 将 50 只筛选合格小鼠随机分为模型组、GG 低组、GG 中组、GG 高组和 空白对照组,除空白对照组外,其余四组各鼠均皮下注射利血平 1. 0mg/kg,连续注射 10 天,观察葛根通络颗粒 对小鼠脑内 5 - HT 含量的影响。结果 痛阈值: 给药前及给药 0. 5h 后四组小鼠痛阈值无统计学意义( P≥ 0. 05) ; GG 高组给药后 1h、1. 5h、2h 与模型组相比,痛阈值明显提高,差异有统计学意义( 分别为 P < 0. 01,P < 0. 01,P < 0. 05) ; GG 中组给药后 1. 5h、2h 与模型组相比,痛阈值明显提高,差异有统计学意义( P < 0. 05) ; GG 低 组给药后各个时间点与模型组相比痛阈值差异无统计学意义( P≥0. 05) 。扭体次数: GG 低、中、高剂量组、阿司 匹林组与模型组相比,都能显著减少冰醋酸导致小鼠扭体次数,差异有统计学意义( P < 0. 05) 。脑内 5 - HT 含 量: 模型组小鼠脑内递质 5 - HT 较正常小鼠明显降低( P < 0. 05) ,GG 高组对小鼠脑内的递质 5 - HT 有明显的 升高作用,与模型组比较差异有统计学意义( P < 0. 05) ,GG 中、GG 低组 5 - HT 的含量较模型组也有一定程度 的升高,但差异无统计学意义( P≥0. 05) 。结论 葛根通络颗粒有一定的镇痛作用,机制可能与升高脑内 5 - 羟 色胺水平有关。
Abstract:
To observe the analgesic effect and the mechanism of pueraria Tongluo granules. Method: 48 eligible mice were randomly divided into: model group,puerarin Tongluo low,medium and high dose groups ( 3. 0,6. 0, 12. 0g · kg- 1) ,in terms of crude drug,hereinafter referred to as GG low group,GG Group,GG high group) . Themodel group was treated with 0. 5% CMC - Na suspension ( 0. 2mL /10g) ,and hot plate method was used to observe the change of pain threshold of mice. 50 eligible mice were randomly divided into model group,GG low group,GG Group, GG high group and aspirin group( 0. 2g / kg) . 0. 5% CMC - Na suspension ( 0. 2mL / 10g) was given to the model group. The mice in each group were injected intraperitoneally with 0. 6% acetic acid ( HAC) at 0. 1mL / 10g one hour after the administration. Mice writhing reaction times were recorded and the inhibition rate was calculated. 50 eligible mice were randomly divided into model group,GG low group,GG group,GG high group and blank control group,except the blank control group,the other four groups were subcutaneously injected reserpine 1. 0mg / kg for continuous 10 days,the effect of pueraria granules on 5 - HT content in mouse brain was observed. Result: Pain threshold: the pain thresholds of the four groups of mice before treatment and 0. 5h after administration were not statistically significant ( P≥0. 05) . Compared with the model group,the pain threshold of the GG high group was significantly increased at 1h,1. 5h and 2h ( P < 0. 01, P < 0. 01,P < 0. 05,respectively) . Compared with the model group,the pain threshold of the GG group was significant- ly increased at 1. 5h and 2h after administration ( P < 0. 05) ,the difference was statistically significant ( P < 0. 05) . Compared with the model group at each time point,the difference of pain threshold of the GG low group was not statisti- cally significant ( P≥0. 05) . Writhing times: Compared with the model group,the GG low,medium and high dose group and aspirin group could significantly reduce the writhing times of mice induced by glacial acetic acid,and the difference was statistically significant ( P < 0. 05) . 5 - HT content in brain: The neurotransmitter 5 - HT in the mice of the model group was significantly lower than that in the normal mice ( P < 0. 05) ,GG high group showed a significant increase in 5 - DT and the difference had statistical significance( P < 0. 05) compared with the model group,5 - DT contents in GG and GG high groups were also increased to some extent while the difference had no statistical significance ( P≥0. 05) .
