摘要:目的 观察黄连温胆汤(HLWDD)对精神分裂症样模型小鼠行为的影响。方法 6周龄雄性KM小鼠,随机分为对照组、模型组、奥氮平(OLZ)组和HLWDD高、中、低剂量(HLWDD-H,HLWDD-M,HLWDD-L)组,每组8只。对照组腹腔注射生理盐水,其余三组注射地西平马来酸盐溶液(MK-801)建立精神分裂症(schizophrenia,SCZ)模型;OLZ、HLWDD组同时灌胃对应药物。末次给药后行旷场、悬尾和水迷宫实验。结果 与对照组相比,模型组体重未见显著差异;旷场活动距离明显增加(P<0.05),中心区域活动时间明显降低;悬尾时不动时间明显增加(P<0.05);水迷宫逃避潜伏期显著增加(第1~4 d,P<0.05;第5 d,P<0.01),目标象限活动时间(P<0.05)及穿越平台次数(P<0.01)均显著降低。与模型组相比,OLZ组体重在给药3~5 w显著增加(P<0.05),HLWDD组未见显著差异;旷场实验中,OLZ组活动距离明显降低(P<0.05),中心区域活动时间明显增加,HLWDD组的活动距离未见差异,仅中心区域活动时间明显增加(P<0.05);悬尾实验中,仅HLWDD-H,HLWDD-M组不动时间显著降低(P<0.05);水迷宫实验中,OLZ组逃避潜伏期在第2~5 d显著降低(P<0.05),穿越平台次数显著增加(P<0.01)。HLWDD-H组逃避潜伏期在第4~5 d显著降低(P<0.05),目标象限活动时间(P<0.05)及穿越平台次数(P<0.01)均显著增加。结论 HLWDD可改善SCZ小鼠的阴性及认知行为,且无增重副作用。本研究为HLWDD治疗SCZ提供了新依据。
Abstract:
Abstract: Objective To observe the effect of Huanglian Wendan decoction (HLWDD) on the behavior of schizophrenia like model mice.Methods 6-week-old male KM mice were randomly divided into a control group,a model group,an olanzapine (OLZ) group,and HLWDD high,medium,and low dose (HLWDD-H,HLWDD-M,HLWDD-L) groups,with 8 mice in each group.The control group was injected with physiological saline intraperitoneally,while the other three groups were injected with MK-801 to establish an SCZ model;OLZ and HLWDD groups were given corresponding drugs by gavage simultaneously.After the last administration,open field,tail suspension,and water maze experiments were conducted.Results Compared with the control group,there was no significant difference in body weight between the model group and the control group;The distance of open field activities increased significantly (P<0.05),and the activity time in the central area decreased significantly;The immobility time during tail suspension significantly increased (P<0.05);The latency period of water maze escape significantly increased (1-4 days,P<0.05;5 days,P<0.01),while the target quadrant activity time (P<0.05) and the number of platform crossings (P<0.01) were significantly reduced.Compared with the model group,the weight of the OLZ group significantly increased (P<0.05) after 3-5 weeks of administration,while there was no significant difference observed in the HLWDD group;In the open field experiment,the activity distance of the OLZ group was significantly reduced (P<0.05),and the activity time in the central area was significantly increased.There was no difference in the activity distance of the HLWDD group,only the activity time in the central area was significantly increased (P<0.05);In the tail suspension experiment,only the HLWDD-H and HLWDD-M groups showed a significant decrease in immobility time (P<0.05);In the water maze experiment,the escape latency of the OLZ group was significantly reduced from day 2 to day 5 (P<0.05),and the number of platform crossings was significantly increased (P<0.01).The evasion latency of the HLWDD-H group significantly decreased on days 4-5 (P<0.05),while the target quadrant activity time (P<0.05) and the number of platform crossings (P<0.01) significantly increased.Conclusion HLWDD can improve negative and cognitive behavior in SCZ mice without any weight gain side effects.This study provides new evidence for the treatment of SCZ with HLWDD.
