Effect of "Sniffing Three Needles" on Expression of iNOS and Phosphorylated tau Protein in Hippocampus of AD Mice

- To observe the effect of " sniffing three needles" on expression of iNOS and phosphorylated tau protein in hippocampus of AD mice and its mechanism of action in treating AD． Method: 40 AD mice were randomly divided into model group，sniffing three － needle group，three － needle olfactory nerve transection? group and donepezil hydrochloride group，and 10 littermates were used as control group． The control group and the model group were subjected to the same degree was kept for 20 minutes． The donepezil hydrochloride group was intragastrically administered with donepezil hydrochloride 5 times per week for a total of 4 weeks，with an interval of 2 days． After the intervention， the expression of p － tau Thr181 protein in hippocampus of mice was detected by Western blot，and the expression of iNOS protein in hippocampus of mice was detected by immunohistochemistry． Result: Compared with the control group，the expression of p － tau Thr181 and iNOS protein in the hippocampus of mice in the model group increased significantly ( P ＜0． 01) ． Compared with the model group， the sniffing three － needle stimulation and donepezil perfusion could significantly reduce the expression of p － tau Thr181 and iNOS protein in the hippocampus of mice ( P ＜ 0． 05) ，and three － needle olfactory nerve transection group had no significant effect on it ( P ＞0． 05) ． There was no significant difference between the sniffing three － needle group anddonepezil hydrochloride group ( P ＞0． 05) ． Conclusion: Based on the integrity of olfactory pathway，"sniffing three needles" may reduce the expression of inflammatory factor iNOS and phosphorylated tau protein in hippocampus of AD mice． of grasping at the same time every day without intervention，the sniffing three － needle group was stimulated by electroacupuncture on three intervention points of“Yintang”and“Yingxiang”in the two sides，and the needle
